Advanced Optical Microscopy Techniques for Multi-scale Biological Studies

Abstract

The blooming development of microscopy techniques has shed lights on many long-term unsolved mysteries in life science. The key points to implement a fruitful imaging-based approach to decipher a biological question encompass the selection of a suitable imaging platform, the ability to modify the standardized hardware and/or software to adapt to the specific experimental requirements, and the proper sample preparation such as mounting and labeling methods. In this talk, I will introduce the basic principles and the characterizations, including the pros and cons, of various advanced optical microscopy techniques, including light-sheet microscopy (LSM), lattice light-sheet microscopy (LLSM), and stimulated emission depletion microscopy (STED), along with the demonstrations of several biological applications I have been working on at different observation scales. The multi-scale and multi-dimensional fluorescence imaging strategies promise to provide insight and possible solutions to tackle the challenges associated with the critical biomedical questions.
